- W2536818761 abstract "The amyloid beta (Aβ) 42:40 ratio has been proposed to be a more stable AD biomarker by (i) correcting for differences in amyloidogenic processing, and/or (ii) abrogating pre-analytical confounders that could give falsely low Aβ42 concentrations. We tested the latter proposal under two pre-analytical conditions known to modify measurable Aβ concentration. CSF Aβ38, 40 and 42 peptide concentrations were analysed from two aspects of storage and handling procedure in cerebrospinal fluid (CSF) and cell culture media (CM). The first studied the effect of storage volume (ranging 100μL to 1000μL) in CSF (n = 8) and CM (n = 6). The second studied the effect of serial transfer between tubes (0-7 transfers) in CSF (n = 7) and CM (n = 5). Assays were run on a Meso Scale Discovery SECTOR 6000. Replicating previous findings, measurable Aβ38, 40 and 42 peptide concentrations decreased significantly with decreased storage volume and over sequential transfers between tubes in both CSF and CM. Ratios between Aβ42:38, Aβ42:40 demonstrated that the relative concentration loss of Aβ42 was exaggerated compared to the shorter peptides under both treatments in both CSF and CM (all comparisons: p = <0.01). Ratios of Aβ40:38 demonstrated that relative concentration loss of Aβ40 tended to be greater, though not to the degree seen in Aβ42. Results suggest adsorption to the tube surface is concentration dependent. Data was analysed using mixed models in R. The adsorbance propensity of longer Aβ peptides (such as Aβ42) to polypropylene surfaces is significantly greater than shorter peptides (Aβ40 and Aβ38 respectively). Consequently, Aβ peptide ratios may become compromised during the aliquoting process. The Aβ42:40 ratio is not a more stable marker than Aβ42 alone from this particular pre-analytical aspect. However, improved diagnostic accuracy of the ratio over CSF Aβ42 is, by implication, more likely to relate to biological factors (correcting for low vs. high producers) rather than peptide loss in the tubes." @default.
