- W2553589721 abstract "Cigarette smoke(CS) plays an important role in the small airway remodeling in COPD, however, its mechanism has not been fully explained. Recently, the elevated plasma Serotonin (5-hydroxytryptamine; 5-HT) levels were found in COPD patients. 5-HT combined with its two receptors, 5-HT2A and 5-HT2B, is known to increase proliferation and collagen synthesis by fibroblasts, thus, plays a role in lung fibrosis. While the role of 5-HT and 5-HT(2A/2B)receptors in airway remodeling induced by CS is still unknown. Ketanserin(5-HT2A receptors inhibitor),RS-127445(5-HT2B receptors inhibitor),quercetin (a natural 5-HT(2A/2B)receptors inhibitor) and saline were administered intraperitoneally to BABL/C mice separately, which were then exposed to CS for 6 and 12 weeks. The expression of serotonin receptors 5-HT2A and 5-HT2B increased in the lung after exposure to CS for 6 and 12 weeks, as assessed by immunohistochemistry. The accumulation of macrophages and neutrophils and increases in IL-1β and TNF-a concentration in BALF and lung tissue were inhibited by Ketanserin,RS-127445 and quercetin intervention. Ketanserin,RS-127445 and quercetin pretreatment suppressed CS-induced goblet cell hyperplasia in mice after exposure to CS for 6 and 12 weeks proved by AB/PAS staining.Furthermore, the fibrosis surrounding the airway increased in the lung after exposure to CS for 12 weeks, not 6 weeks proved by elastica-Masson9s staining. Ketanserin,RS-127445 and quercetin pretreatment significantly attenuated the thickening and collagen deposition of the small airway. Our results suggest that CS-induced small airway remodeling is partially mediated by the 5-HT and 5-HT(2A/2B)receptors." @default.
