In proceedings of the first international conference on stable isotopes in chemistry, biology, and medicine. Levels of 7.81, 18.46, and 26.02 mole% D/sub 2/O were given successively to Japanese quail and gross biological effects were measured. The most significant effect was a decrease in egg frequency during the period of 26.02 mole% D/sub 2/O administration. The maximum D concentration in the water of the egg was reached within 96 to 100 hrs after the initial administration and it was 6.90, 14.55, and 20.15 mole% respectively during the 3 periods of D/sub 2/ O administration. These values implied 21 to 22% of the water in the egg was water derived from metabolism, during the latter two periods, compared to 12% when the lowest level of deuterium was fed. The amino acids were isolated from the egg white proteins obtained during the 18.46 mole% D/sub 2/O feeding, and analyzed for deuterium content by mass spectrometry of their respective dansyl derivatives. As an example, the results for alanine indicated a mole per cent distribution of 56 plus or minus 1.4 unlabeled, 34.6 plus or minusmore » 1.1 monolabeled, 8.3 plus or minus 0.2 doubly labeled, and 1.0 plus or minus 0.2 triply labeled molecular species. Initial interpretation of these results implies that the quail synthesizes all of the alanine necessary for egg white protein synthesis, utilizing no dietary alanine. A final interpretation of metabolic pathways involved is dependent on knowledge of distribution of label within the molecule and degree and distribution of label in carbohydrate percursors.
