- W2588366957 abstract "Regulation of cell division in Streptomyces venezuelae-Popular science summary-Cell division is essential for bacteria to reproduce. This process involves FtsZ, an important and highly conserved cell division protein in bacteria. FtsZ plays a major role in the division mechanism by forming ring-like structure at the site of division, recruits and organizes other cell division proteins at the division site and contributes to membrane constriction. There is a rising problem with increasing antibiotic resistance among pathogenic bacteria. By gaining more knowledge about essential cell functions in bacteria, such as cell division, one could find new targets for antibiotics. Streptomyces have a huge industrial and medical importance because it is the major antibiotics-producing genus, and it is closely related to important pathogens like Mycobacterium tuberculosis. Therefore, its regulation mechanisms of cell division are of interest. Streptomyces venezuelae shows a very good potential to become a powerful actinobacterial model organism in general and especially study of cell division because of its unique growth behavior and all the tools and data available. The main objective of this study is to investigate in detail how FtsZ assembly is regulated during sporulation in Streptomyces (figure. 1) and to exploit the promising features of Streptomyces venezuelae to establish procedures and tools for investigation of cell division. In order to visualize FtsZ during cell division, several S. venezuelae with FtsZ-fluorescent protein fusions were constructed and assessed. Then, several S. venezuelae cell division mutants were constructed and analyzed using different molecular biology tools. The results show that some mutants have no obvious defects in cell division and produce spores of similar size of as the wildtype strain. Due to defects in division, other mutants produce longer and more irregular spores compared to the wildtype. An FtsZ-YPet (yellow fluorescent protein) fusion protein was constructed and used as a tool to visualize and compare the FtsZ assembly between wildtype and the mutants. In order to better visualize the protein assembly that orchestrate division, during this research, fluorescence time-lapse microscopy for S. venezuelae was established for the first time. Together with the fusion protein constructions and creation and analysis of the cell division mutants, some more insight of genes that are required for proper control of cell vision in S. venezuelae is revealed.
