• W2588718389 abstract "The physiological action of luteinizing hormone releasing hormone (LHRH) depends on pulsatile patterns of secretion. Continuous stimulation of the pituitary by LHRH, which may be achieved either by high doses or by constant infusion, results in (1) desensitization of the pituitary gland, (2) failure of gonadotropin secretion, (3) involution of the gonads and (4) regression of organs, structures and functions that depend on gonadal steroids for maintenance. This paradoxical antifertility effect of LHRH can be mimicked by the administration of antagonistic analogs of LHRH, which block the effect of the parent hormone at the level of the pituitary receptors. Peripheral effects of LHRH acting directly on the gonads were described, but their physiological significance remains to be elucidated. In addition to these antagonistic analogs, there are a number of agonistic analogs that are significantly more potent than their parent when assayed for their ability to release LH. The increased potency of at least some of these appears to result from amino acid substitutions that resist metabolic degradation and permit longer exposure of the LHRH receptors to the analog. LHRH and its analogs have a broad range of therapeutic applications. These segregate into the effects of LHRH and agonistic analogs, which have both stimulatory and paradoxical inhibitory actions, and the effects of antagonistic analogs." @default.
