- W2594787110 abstract "A critical requirement for 3D cell culture is to be able to recreate the cellular microenvironment with a physiologically relevant matrix that is cell specific and tunable. In addition, the matrix must be optically transparent, easy to use and amenable to downstream microscopic and immunohistochemical analyses required for high-content analysis. We describe one such hydrogel matrix platform, HyStem®, a hyaluronan-based hydrogel that contains requisite cellular attachment sites, and that can be easily modified in terms of composition and form. HyStem is composed of three components: thiolated hyaluronan, thiolated porcine gelatin and a diacrylated, thiol-reactive crosslinker polyethylene glycol diacrylate which links the two together in approximately 20 minutes. The thiol-based chemistry allows the user to chemically conjugate a variety of biological moieties into the hydrogel (peptides, proteins, antibodies) and in situ gelation allows customisation of form (coatings, beds, discs, microspheres) as a hydrogel, film or sponge. An example of its flexibility in developing a high-content, medium-throughput assay is described, illustrating the ease with which a 3D assay using standard drug discovery equipment can be employed to prepare and analyse the resulting 3D cellular constructs." @default.
