- W2594912864 abstract "Summary Oxygen uptake experiments with tyrosinase from wheat, tyrosine, DOPA and chlorogenic acid on the presence and absence of bisulfite were conducted to determine the possible mechanism of the action of bisulfite in the inhibition of the enzymatic darkening of the dough prepared from the whole meal flour of Mexican varieties of wheat. The presence of bisulfite with tyrosine resulted in an induction period before the oxidation of tyrosine began. The rate of oxidation of tryosine was decreased with successive increase in the concentration of bisulfite. DOPA and chlorogenic acid oxidations were also inhibited in presence of bisulfite but the effects were not so severe as in case of tyrosine. Tyrosine plus chlorogenic acid and DOPA plus chlorogenic acid consumed much more oxygen than any of the three substrates alone. Addition of chlorogenic acid to the reaction mixture containing bisulfite and any other substrate, reduced the effective inhibitor concentration to practically non-inhibitory levels. From the results reported here, it appears that the bisulfite does not have any effect on the active site of the enzyme, rather it combines with the quinones produced in the reaction mixture by the oxidation of the substrates commonly involved in this type of enzymatic darkening, to give addition products which further inhibit the enzymatic oxidation by competitive mechanism. Higher concentrations of bisulfite required to inhibit such type of enzymatic darkening were explained on the basis of interactions between chlorogenic acid, tyrosine and bisulfite in these enzymic oxidations." @default.
