- W2596431415 abstract "A mid-cycle surge of luteinizing hormone (LH) activates receptors on granulosa cells of preovulatory (PO) ovarian follicles to promote progesterone biosynthesis, ovulation, and luteinization. These acute changes in follicle function are likely dependent on dynamic changes in the granulosa cell cytoskeleton. We have recently shown that microtubule associated protein (MAP) 2D is expressed in rat PO granulosa cells and that LH receptor-mediated dephosphorylation of MAP2D at Thr256/Thr259 occurs through interaction with a signaling complex involving glycogen synthase kinase (GSK) 3 beta, the protein phosphatase (PP) 2A subunit B56 delta, and cAMP-dependent protein kinase (PKA). MAP2 family proteins have been extensively characterized in neuronal systems with a particular focus on their binding to microtubules, actin microfilaments, and intermediate filaments. Here we describe direct binding and colocalization of granulosa cell MAP2D and vimentin intermediate filaments. This binding is reduced by phosphorylation of MAP2D by GSK3 beta at sites including Thr256/Thr259. We present data supporting the hypothesis that LH receptor-mediated MAP2D phospho-regulation coordinates redistribution of MAP2D and associated PKA-signaling complex to vimentin filaments, allowing for PKA-dependent vimentin phosphorylation and reorganization of the vimentin cytoskeleton. We propose that such vimentin remodeling may facilitate the biosynthesis of progesterone that is obligatory for ovulation. Supported by RO1 HD 046955 to MHD." @default.
