- W2600029101 abstract "糖質加水分解酵素ファミリー31に属するα-グルコシダーゼは，基質の重合度に対する特異性が酵素起源によって様々である．本研究では植物α-グルコシダーゼの長鎖基質認識機構を明らかにすることを目的とし，新規の長鎖阻害剤の酵素合成およびそれらとテンサイα-グルコシダーゼとの複合体X線結晶構造解析を行った．長鎖阻害剤合成では，D-酵素による不均化反応を利用することで，アカルボース(AC4，擬似四糖) を伸長させた擬似五糖～擬似十糖(AC5～AC10) を得た．速度論解析の結果，AC5～AC7はAC4と同様にテンサイα-グルコシダーゼの加水分解反応における遷移状態を模倣していることが示された．また，テンサイα-グルコシダーゼとAC8との複合体構造解析の結果，活性ポケットから離れた位置ではAC8が分子内水素結合を形成した安定ならせん構造をとっていた．以上の結果から，テンサイα-グルコシダーゼは安定ならせん構造の基質との結合に適したサブサイト構造を有しており，安定ならせん構造の基質との結合がKmの低下および遷移状態の安定化につながっていると考えられた．" @default.
