• W2604990665 abstract "Microsatellites or simple sequence repeats (SSRs) markers have been widely and efficiently used for creating barley linkage maps. Because of their high level of polymorphism and their abundance and low costs of assay, SSRs markers have been chosen for the creation of a barley linkage map based on a new doubled haploid population. The population of seventy seven doubled haploid lines were derived from a cross between Rihane and MK1a using the Hordeum bulbosum method. Rihane is a six-rowed feed barley cultivar extensively cultivated in North Africa and MK1a is a six-rowed barley line derived from a cross between Tunisian landraces. In order to screen for polymorphism between the parents one hundred and eighty nine SSRs markers were used. Thirty percent of them were shown to be polymorphic on silver stained denaturating polyacrylamide gels. The segregation of fifty two loci in the doubled haploid population was studied and a linkage map was constructed. A deviation to the 1:1 expected segregation was observed for twelve (23.5%) microsatellite loci. They are probably linked to a selected group of genes during the process of haploid production. The linkage map covering 317.4 cM of the genome contained thirty two SSRs markers placed on nine linkage groups. Comparisons with published maps showed a high level of co-linearity, although some discrepancies in loci order exist between closely linked markers." @default.
