- W2605607114 abstract "Purpose: To explore another way for disappearance subchondral bone cyst of Osteoarthritis (OA), which was under treatment by using some drugs focused on improving the mineral density so far. Subchondral bone supports overlying cartilage biomechanically. It has been shown that subchondral bone cyst have great relationship with early osteoarthritis. Methods: A new regenerative treatment method was raised in this study: porous gelatin-chitosan-hydroxyapatite micro-cryogels which could be directly injected can induce the regeneration of diseased bone tissue and the recovery of cyst-like bone defect in vivo. Meanwhile the chitosan-hydroxyapatite compound could also act as a key factor to adjust bone regeneration and reduce the immune rejection. Human infrapatellar fat pad derived stem cells (FPSCs) cultured with different scaffolds in induced medium were used to testify the osteoinductivity of gelatin-chitosan-hydroxyapatite micro-cryogels. Results: SEM images of the microcryogels showed good interconnected and macroporous structures. The X-ray imaging, histology staining in vivo showed no side effect such as thrombosis and severe inflammatory reaction. Human infrapatellar fat pad derived stem cells (FPSCs) cultured with different scaffolds in induced medium showed that the osteoinductivity of gelatin-chitosan-hydroxyapatite micro-cryogels comparing with gelatin micro-cryogels. After 8 weeks’ injection in rabbit subchondral bone cyst-like defect model, the gelatin-chitosan-hydroxyapatite micro-cryogels group showed better regeneration in Micro-CT imaging, H&E staining and Masson trichrome staining. Conclusions: Our results provide in-depth demonstration that the injectable gelatin-chitosan- hydroxyapatite micro-cryogels, a leak-proof cell gathering system, can enhance bone tissue regenerative therapy of subchondral bone cyst-like defects in rabbit models." @default.
