- W2624887470 abstract "Neurological symptoms of patients suffering from neurodegenerative diseases such as Alzheimer´s dementia often worsen during infections. An experimental approach to this clinical observation was performed in vivo and in vitro. We studied the effects on a mouse model of Alzheimer´s dementia from recurrent systemic infections with the most frequent respiratory pathogen of adults: Streptococcus pneumoniae. Transgene mice over expressing the Aβ-Precursor-Protein (TG2576) were repeatedly challenged with vital Streptococcus pneumoniae Type 3 and treated with the bactericidal antibiotic Ceftriaxon for three days. Inoculation caused an inflammatory response in the CNS with an increase of interleukin-6 concentration in brain homogenates. Motor performance was controlled by the tight rope test and the rotarod test and did not show any deficits. Spatial learning, memory and learning deficits were assessed using the Morris water maze. Neither onset nor course of Alzheimer´s dementia was affected by the recurrent systemic infections. Plaques were dyed with thioflavin S and showed no size differences between infected and unifected mice. Detergent-soluble and detergent-insoluble Aβ-fragments 1-37, 1-38, 1-39, 1-49 and 1-42 from brain homogenates were detected via westernblot. Concentrations for the detergent-soluble fraction of Aβ 1-40 and Aβ 1-42 were also measured via the more sensitive ELISA. Concentrations of Aβ-fragments were not significantly different in brains of infected and uninfected Tg2576 mice. In conclusion, onset and course in a mouse model of Alzheimer´s disease was not influenced by repeated systemic infections with Streptococcus pneumoniae, indicating that the effect of moderately severe acute infections on the course of Alzheimer´s disease may be less pronounced than suspected. In addition we studied the combined effects of Aβ 1-40 and bacterial components on murine or human neuronal-monocytic co-cultures. Although our theory of co-activation was supported by the visual impact, we could not generate a usable statistical result due to the difficulty to quantify the cell damage." @default.
