• W265434549 abstract "An attempt was made to formulate microspheres loaded with Doxycycline for insertion into the periodontal pocket for achieving enhanced local drug concentration for sustained effect, thereby, decreasing systemic side effects. After optimization of a series of process parameters, Doxycycline loaded chitosan microspheres were prepared by emulsion cross-linking technique. The microspheres were subjected to characterization tests such as particle size, recording of particle shape, surface morphology, calculation of percentage yield, entrapment efficiency, ex-vivo mucoadhesion test, swelling extent and ex-vivo drug diffusion through sheep buccal mucosa. The formulated microspheres displayed free flowing properties, were spherical having a mean particle diameter of 81.26 ± 2.33 µm. Increased drug entrapment efficiency was achieved with increased polymer concentration leading to increased particle size. An entrapment efficiency of 84.3 ± 0.77% was observed. The microspheres did sustain the drug release for a period of 8 h. Upon fitment of data to various kinetic models; the drug release was found to occur by matrix diffusion and by non-Fickian mechanism. Select formulations were subjected to stability studies as per ICH guidelines for 90 days. No changes were observed in respect to ex-vivo mucoadhesion, drug diffusion pattern and the drug content remained within permissible limits." @default.
