- W266986639 abstract "The phenomenon of ammonia related growth inhibition has been widely reported. The accumulation of ammonia in culture is a direct result of the chemical and metabolic deamination of glutamine which is normally present in growth medium at concentrations between 2-4mM. We have investigated the effects of ammonia on three cell lines; Baby Hamster Kidney cells (BHK21 C13), McCoy, and Vero cells grown on serum supplemented GMEM in T-flasks or microcarrier cultures. Concentration dependent inhibition of cell growth, by ammonia, was shown to vary between cell lines. In order to reduce ammonia accumulation in these cultures, we have developed a protocol which allows adaptation of cells to glutamine free media. Glutamate or 2-oxoglutarate were substituted for glutamine in these media formulations and resulted in the reduction of accumulated ammonia by 50%. Other effects of adaptation of cells to glutamine free media include altered glucose consumption and lactate production. The implications of this adaptation protocol to the achievement of higher cell productivities in microcarrier cultures will be discussed." @default.
