En este trabajo, caracterizamos las secuencias de la mitad 5’ de la RNC de los haplotipos identificados en las colonias de anidacion del suroeste de Cuba de Chelonia mydas y Caretta caretta, teniendo en cuenta el tipo y sentido de sustitucion nucleotidica, respecto al haplotipo ancestral y los centros de diversificacion determinados dentro de cada linaje. Los haplotipos de C. mydas pertenecieron a un solo linaje, mientras que en C. caretta a dos. La mayoria de las sustituciones intraespecificas fueron transiciones, con balance similar entre purinas y pirimidinas, asi como entre los sentidos de las sustituciones en relacion al camino mas parsimonioso respecto al ancestro. El mayor porcentaje de estas sustituciones representaron cambios diagnosticos para C. caretta respecto a C. mydas. Los porcentajes equitativos de transiciones entre purinas y pirimidinas, concuerdan con la abundancia de A y T, y con la predisposicion a la conversion de CpG por el alto porcentaje de metilacion de C. Chelonia mydas presenta una distribucion de mutaciones similar entre los linajes de la cuenca atlantica a diferencia de C. caretta, porque los de esta ultima no comparten un ancestro comun. Por consiguiente, entre estos linajes pueden ocurrir homoplasias y fijarse un mayor numero de mutaciones, apareciendo transversiones e indels diagnosticos de linajes en la medida en que aumentan las distancias evolutivas entre estos. Palabras claves: mtDNA; mutacion; Caretta caretta, Chelonia mydas; ASW, Cuba Abstract Inside mitochondrial DNA (mtDNA), Non Coding Region (NCR) has been reported to have the fastest intragene substitution rate. We considered the type and direction of nucleotide substitution regarding the ancestor haplotype and the diversification centers inside each lineage. sequences of the 5' half from NCR were characterized in identified haplotypes from southwestern Cuba rookeries of Chelonia mydas and Caretta caretta. C. mydas haplotypes only belonged to one lineage, while C. caretta’s belonged to two lineages. Most of the intraspecific substitutions were transitions, with similar balance between purines and pyrimidines, as well as among the substitutions directions in relation to the most parsimonious way regarding the ancestor. A higher percentage of these substitutions represented diagnostic changes for C. caretta compared to C. mydas. Equitatives percentages of transitions between purines and pyrimidines agreed both with the abundance of A and T, and with the bias to the conversion of CpG and, due to the high percentage of metilation of C. C. mydas, showed a similar distribution of mutations among Atlantic basin lineages. On the contrary, C. caretta did not showed the same behavior because their lineages did not share a common ancestor. Consequently, homoplasies may occur among these lineages and a higher number of mutations may be fixed, appearing then lineages´s diagnostic transversions and indels while their evolutionary distances increases. mtDNA; mutations; Caretta caretta, Chelonia mydas; ASW, Cuba" @default.
