• W2743681498 abstract "Abstract Objectives In the past years, there have been increases in S. aureus infections, intensifying the demand for new and effective drugs. Apparently, old drugs such as statins have shown unexpected pleiotropic effects, and may be exploited or repurposed to meet new demands on antimicrobial therapy. In this study, the antibacterial activity of atorvastatin and possible interactions with ampicillin were evaluated by determining the minimum inhibitory concentration (MIC), synergism, and biofilm inhibition of these drugs against S. aureus ATCC 29213. Methods In this study, atorvastatin and ampicillin, alone or in combination, were tested for antibacterial property against S. aureus using the MIC assay, and tested for synergism via the checkerboard microdilution assay. Both drugs were then tested against biofilm formation to check its relevance to sessile bacterial states. Results Atorvastatin did not display anti-bacterial activity consistent with previous reports, but synergistically inhibited planktonic S. aureus growth by lowering the MIC of ampicillin by 8-fold. Further, atorvastatin abrogated biofilm formation in 48h-grown S. aureus − implicative of its role in chronic infection, but lost this property when co-administered with ampicillin in a ratio of 1:1. Conclusion These suggest that atorvastatin and ampicillin may in fact be targeting a common bacterial pathway integral to biofilm formation, and this warrants further verification on the mechanisms involved as well as on the identification of novel enzyme off-targets that could potentially impact the treatment of S. aureus infections." @default.
