- W2744164170 abstract "The integrative properties of neurons under in-vivo-like conditions can be studied in a controlled way through the injection in the cell of a fast fluctuating current depending on the cell's Vm (dynamic-clamp). This “conductance noise” mimicks the stochastic activation of thousands of synapses driven by an intact cortical network. In the many cases when two electrodes cannot be used for dynamic-clamp (adult slices, in vivo), passing current and recording the cell's Vm through a single high-resistance electrode introduces severe artefacts. Classical solutions to this problem either rely on simplified assumptions and cannot remove all of the electrode's contribution to the signal (“bridge mode”: the electrode is treated as a simple RC circuit), or severely reduce the sampling frequency and distort the injected current by alternating current injection and Vm recording (DCC: discontinuous current-clamp). We have developed a new, active on-line electrode compensation method (AEC) which allows faithful recording in continuous mode, with sampling frequency only limited by hardware constraints. The filter of the electrode, modeled as a general linear circuit, is identified by injecting a short period of white noise into the cell, after which the recording is compensated online by digital convolution. Here we present a systematical comparison of current and conductance injection with AEC vs. DCC, both in cortical neurons in vitro and in models. The higher temporal resolution of AEC allows to record responses to short pulses that better match analytical predictions, to explore the shape of fast events such as spikes, to obtain a higher reliability in spike counts and timings during repeated injections of frozen conductance noise, and to reduce aliasing noise. To conclude, AEC extends the range of issues that can now be addressed about in-vivo like cortical states using the single-electrode dynamic-clamp." @default.
