The innovations that drive these techniques are a combination of switchable fluorophores and clever image analysis algorithms that identify single-fluorophore images and then localize the fluorophores. The performance of these algorithms affects the quality and resolution of the reconstructed image through two mechanisms. First, algorithm performance determines the maximum precision with which the fluorophores can be localized. Second, algorithm performance determines the fraction of fluorophores that are successfully identified for analysis. The proper identification of single-fluorophore images is necessary for acquiring sufficient spatial information, and the proper rejection of multi-fluorophore images (or, alternately, their identification for fitting to a multi-fluorophore model) is necessary for assuring image quality. Together, these considerations dictate the maximum achievable experimental speed, by determining the maximum fraction of fluorophores that can be activated in a given frame." @default.
