- W2750920912 abstract "5198 The incidence of prostate carcinoma in the Indian population is very low compared to that of the Western Hemisphere population. Prostate cancer is the 10th most common malignancy among Indians. On the other hand it ranks fourth most common malignancy in men worldwide and is the second most commonly diagnosed cancer in American men. This low incidence of prostate cancer might be attributed to consumption of various spices in the Indian diet. One such spice is turmeric (Ccurcuma longa) in which curcumin is the major active principle. We have previously demonstrated that Curcumin is a potent inhibitor of prostate tumor growth and sensitizes to the effects of radiation therapy. Sequel to our previous studies, in this study, curcumin showed a dose dependent decrease in survival of LNCaP/LN3 (p53 wild type) and DU145 (p53 mutant) cells with IC50 values of 3μM and 5μM respectively as assessed by clonogenic cell survival assay, indicating that p53 might play a role in curcumin mediated toxicity. One of the most aggressive features of prostate cancer is overexpression bcl-2 protein associated with or without the inactivation of the p53 gene that together contributes to a more radioresistant phenotype. Hence, to mimic those conditions, we selected transfected PC-3 cell line that is null for p53 function with vector alone (PC-3/Vector) or full-length cDNA of Bcl-2 (PC-3/CMV-Bcl2). Curcumin inhibited the growth of PC-3/Vector cells with an IC50 value of 1μM and PC-3/CMV-Bcl2 with an IC50 of 3.5μM.
