- W2756285151 abstract "Abstract Autophagy is a catabolic process that involves the packaging of intracellular proteins and organelles into membrane vesicles called autophagosomes, with the autophagosomes subsequently delivered to lysosomes for degradation. The assembly and maturation of autophagosomes is regulated by interactions with different membrane compartments, and recent studies have highlighted critical roles of intracellular trafficking machinery in autophagy regulation. This chapter addresses the role of a key vesicle fusion mediator, soluble N -ethylmaleimide sensitive factor–attachment protein alpha (αSNAP), in autophagy. We discuss recent data that highlight αSNAP as a dual regulator of the autophagic flux in human epithelial cells. The most unusual and previously unanticipated activity of αSNAP involves inhibition of autophagy, which does not depend on the canonical functional partner of αSNAP, N -ethylmaleimide sensitive factor. This noncanonical function of αSNAP is related to its role in the regulating Golgi structure, thereby limiting delivery of Golgi-derived vesicles into the autophagic pathway. This role of αSNAP in autophagy regulation is discussed in a broader context of recently discovered new binding partners of this adaptor protein and its novel roles in the regulation of epithelial homeostasis." @default.
