- W2760759909 abstract "There is increasing evidence to support the role of cancer stem cells (CSCs) in cancer development, chemoresistance, and relapse; resulting in poor prognosis and high mortality rate of cancer. The tumor microenvironment has been implicated to have an influence on cancer cells in many ways, where cell signaling molecules and mediators including proinflammatory cytokines and reactive species are frequently upregulated. Among them, the concentrations of nitric oxide (NO), a reactive nitrogen species synthesized by many cell types, are found to be upregulated in many cancer microenvironments, and elevated NO levels have been linked to advanced stage and poor survival of cancer patients. A plethora of studies demonstrate the oncogenic activity of NO in tumor progression; however, the underlying mechanisms remain poorly understood. This chapter summarizes and discusses the effects of NO on tumorigenesis and metastasis with a focus on CSC regulation and its effect on tumor progression. NO in the tumor environment appears to have a significant impact on the metastatic behavior of CSCs by affecting hallmarks of cancer including unlimited cell proliferation, evasion of apoptosis, increased cell migration and invasion, and capability of initiating tumor formation. Recent studies have indicated that NO-mediated CSC phenotype occurs through multiple pathways such as induction of target proteins like caveolin-1, expression of CSC markers, and via epithelial-mesenchymal transition. We discuss the emerging evidence documenting the significance of NO in the regulation of CSC phenotype that appears to be of the plausible mechanisms for NO-mediated aggressive behavior of tumor cells. These findings uniquely address the role of NO in CSC-mediated metastasis and can facilitate new drug discovery strategies for cancer treatment." @default.
