- W2765796889 abstract "Physical activity (PA) interventions have successfully increased PA among sedentary older adults. However, increases are generally not sustained, with PA increasing early then declining and returning to sedentary levels after trial completion. Dopamine may play a role in PA adherence as dopaminergic networks regulate factors related to PA in older adults, including cognitive control, physical function and motivation/reward response. PA was measured in initially sedentary older adults in the LIFE study (n=1,373, ages 70–89, 66% female, 23% black) randomly assigned to a PA program of center-based walking 2X/week or a health education (HE) program, both for ≥24 months. Moderate levels of PA were objectively measured by accelerometry as minutes/day at baseline, 6, 12 and 24 months. Single nucleotide polymorphisms (SNPs) were measured in genes that regulate dopamine availability and neurotransmission (DRD1, DRD2, DRD3, COMT). PA levels were compared by SNP at each visit and over time, stratified by race. Interactions of intervention arm with gene polymorphisms was tested. The PA and HE groups had similar health and demographic characteristics, genotype distribution (p range=0.2–0.9), and levels of PA at baseline (p=0.7), but significantly higher PA at follow-up visits (all p<0.05). At 6 months, white older adults in the PA group with the DRD2 met/met allele (indicative of higher dopamine availability) spent significantly more time in moderate PA (25.9±29.5 minutes/day) compared to those with either the val/val (21.6±28.2 minutes/day) or the met/val (20.5±25.0 minutes/day) allele (p=0.01). Results were similar at 12 and 24 months (p=0.02 and 0.01). Longitudinal models confirmed significant interactions between DRD2 and arm and more sustained increases in PA for those with met/met allele. There were no significant differences by DRD2 genotype for black older adults or by allele for any other SNP or in the HE group. Higher dopamine availability may be important for sustained increases in PA in response to lifestyle modifications. While genotype is not a modifiable factor, the mechanistic insights may indicate novel pathways to promote sustained increases in PA in sedentary older adults. Studies to examine the role of dopamine-related pathways for sustained PA adherence in older adults are warranted." @default.
