- W2765981303 abstract "Partial volume (PV) effects in cortical target and reference regions used in a VOI-based SUVR analysis can bias longitudinal results. We investigated the effect of PV change between timepoints on longitudinal SUVR in patients with Alzheimer's disease (AD), Mild Cognitive Impairment (MCI) and Normal Controls (NC). MRI data from 270 ADNI subjects (23 probable AD, 145 MCI, 102 NC) at two timepoints approximately 24 months apart was cross-sectionally segmented using Freesurfer 5.3 to produce grey matter, white matter and CSF tissue classes, according to Müller-Gärtner approach. PV contributions of each tissue type were computed for composite and reference regions. First, we evaluated the effect of longitudinal PV changes on SUVR without accounting for increasing amyloid load. Second, we evaluated combined effect of PV changes and changes in amyloid accumulation between timepoints on SUVR. Aβ status (determined via baseline Florbetapir PET) was used to stratify the analysis into AD Aβ+, MCI Aβ+, NC Aβ+, and NC Aβ- groups. SUVR without PV correction increased between timepoints in all groups, irrespectively of the reference region, with largest increase in NC Aβ+, followed by MCI Aβ+ and AD Aβ+, and smallest in NC Aβ-. When PV changes were considered, SUVR decreased between timepoints in all groups. Largest decrease was observed for SUVR with subcortical eroded WM and cerebellar GM reference regions in AD Aβ+, cerebellar WM in MCI Aβ+, cerebellar GM and whole cerebellum in NC Aβ+ and NC Aβ-. One exception was SUVR normalized to corpus callosum, where SUVR values increased in all groups. When annual % change of amyloid SUVR was combined with change in PV, SUVR increased between timepoints in all groups, with the same increasing trends but lower than the increase observed without PVC. Compensation for partial volume effects in longitudinal SUVR analysis must be treated with caution since it may negatively affect estimated SUVR and it is related to the reference region. Accounting for increasing amyloid load did not completely remove a consistent negative bias for PVC on longitudinal SUVR. Considering small effects of prospective anti-amyloid therapies care must be taken when evaluating factors contributing to SUVR change." @default.
