It has a dual role to provide initial control of the infection and to initiate an adaptive immune response. This chapter discusses the factors in the microenvironment that may regulate and augment the innate immune response. It focuses on the role of neutrophils and macrophages and on the coordinated response that they generate to infection or injury. This response depends on many factors in the environment including oxygen availability, availability and utilization of metabolites, pathogen factors, and host damage signals and is mediated via an array of inflammatory (and anti-inflammatory) mediators. For effective immunity, neutrophils and macrophages must be adapted to function in the hypoxic environments. Both neutrophils and macrophages have the capacity to modify the immune response through the production of a wide range of pro- and anti-inflammatory mediators.
