- W2783169314 abstract "We tested if treatment with the selective serotonin reuptake inhibitor, fluoxetine (FX), restores heart rate variability (HRV) in rats with heart failure. Male Sprague-Dawley rats were subjected to left coronary artery ligation (CAL) under ketamine/xylazine anesthesia (100 mg/kg + 7 mg/kg, im). CAL rats (n=9) with low fractional shortening and sham rats (n=16; 16.8 ± 0.9% vs. 46.3 ± 1.7%, P<0.01) were fit with telemetric probes for electrocardiogram recording. Rats were given FX (20 mg/kg, sc) or vehicle, once daily for 5 wks. Power spectral density of HRV was determined in the low frequency (LF, 0.4-1.0 Hz) and high frequency domains (HF, 1.0-3.0 Hz) before and after propranolol (4 mg/kg) and atropine (2 mg/kg). CAL rats had lower total power (4.8 ± 1.3 vs. 30.2 ± 4.7 ms2, P<0.01) due to a decrease in both LF (1.1 ± 0.3 vs. 6.3 ± 1.2 ms2, P<0.01) and HF (1.5 ± 0.7 vs. 4.1 ± 0.8 ms2, P<0.05). FX lowered LF in sham (4.1 ± 0.8 vs. 8.6 ± 1.9 ms2, P<0.01) and CAL rats (0.6 ± 0.4 vs. 2.1 ± 0.4 ms2, P<0.01) and lowered the LF/HF ratio in CAL rats (0.80 ± 0.22 vs. 2.83 ± 1.12%, P<0.05) without affecting HF in either group. FX reduced the effect of propranolol on LF in CAL (−0.06 ± 0.15 vs. −1.01 ± 0.36 ms2, P<0.05), but not sham rats. Atropine further lowered HF in sham (−60.8 ± 10.6%, P<0.01), but not CAL rats. FX did not affect atropine responses. These data indicate that FX lowers HRV selectively in LF, suggestive of a decrease in sympathetic influence on HRV." @default.
