However, their poor pharmacokinetic properties have hampered their widespread application. To overcome such limitations, various bioengineering strategies seeking to extend the half-lives of biopharmaceutics have been devised. Such strategies involve chemical conjugation or genetic fusion of a therapeutic peptide or protein with synthetic polymers or protein carriers. This chapter summarizes the bioengineering techniques used to create second-generation products that are currently under development or already on the market. Such work has greatly improved pharmacokinetic profiles, often due to fusion of an active protein to the Fc domain of immunoglobulin G, human serum albumin, transferrin, or a carboxy-terminal peptide." @default.
