- W2783285513 abstract "A group of forty seven aerobic actinomycetes including representatives of thirteen genera was assembled. Some of the strains were obtained frwn other workers and the remainder were isolated from local soils using baiting techniques or direct plating methods. Although regular subculturing was the method of choice for maintrft nance of cultures, the effectiveness of a number of storage methods in maintaining viability was investigated* lyophilization and freezing at -20oC were found to be more effective than storage on slopes. Application of the Streptomyces identification methods of Shirling and Gottlieb (1966) indicated that although they are broadly useful for study of strains of other actinomycete genera, there were certain limitations in that some strains were unable to grow on all media. The media were suitable for determination of morphological characteristics of the strains which were capable of growth. The reactions of strains to a range of physiological and biochemical tests were determined for later use in a numerical analysis. Cell-wall analysis results confirmed published data and it was considered that the nature of the diamino acid(s) present in the walls was the most useful taxonomic feature. Analysis of the end products of carbohydrate metabolism by thin layer and gas chromatography indicated than the former. C2 - C5 acids including iso- and normal isomers were produced of which acetic was the principal component. Strains of Dermatophilus congolensis produced chiefly acetic and propionic acids and this appears to set the strains of this species apart from the other actinomycetes studied. Quantitative analyses indicated that under the growth conditions used, only small quantities of supplied glucose appeared as acid end products. Most strains were capable of accumulating various indoles in culture media. These appear to arise from tryptophan supplied in the medium only a small amount of which was converted to indole acetic acid. n" @default.
