Numerous studies have identified beneficial effects from the use of 1,25-dihydroxyvitamin D3 in combination with a second therapeutic agent, which may allow lower doses to be used. An alternative to standard combination therapy is the use of bifunctional molecules where a single species can modulate multiple targets simultaneously. In this review, we discuss the development of hybrids, which combine structural features of 1,25-dihydroxyvitamin D3 with those of several other molecule classes, notably histone deacetylase inhibitors, saponogens, and bisphosphonates, resulting in molecules with improved properties toward models of cancer, osteoporosis, and Alzheimer's disease." @default.
