Dies fuhrt zur Aufnahme der Bakterien via groser Membraneinstulpungen. SfbI-exprimierende Streptokokken umgehen die klassische intrazellulare Wanderroute, die normalerweise die Fusion von Bakterien-enthaltenden Kompartimenten mit Lysosomen und so die Abtotung der Bakterien zur Folge hatte.In dieser Arbeit wurde gezeigt, dass sowohl AroD als auch ProD die SfbI-abhangigen Aufnahme beeinflussen; neben der beschriebenen Aufnahme wurde ein morphologisch unterschiedlicher Mechanismus entdeckt, der mit der Formung von Membranprotrusionen einhergeht. Die Analyse von Co-Inkubationsexperimenten von Zellen mit verschiedenen Proteinkonstrukten, die entweder rekombinant exprimiert fur sich allein oder gekoppelt an Latex beads, oder heterolog exprimiert von S. gordonii vorlagen, ergaben, dass bestimmte AroD Typen oder das Fehlen der ProD die typische SfbI-vermittelte Aufnahme unterdrucken: Integrin-Clustering und Caveolae-Aggregation werden verhindert; stattdessen werden Zytoskelettumordnungen und Signale der klassischen Phagozytose hervorgerufen. Auserdem wird das bakterielle Uberleben von der intrazellularen Route beeinflusst.AroD vermag die SfbI-Konformation zu modulieren, mit Auswirkungen auf die Fn-Bindungseigenschaften und ProD-Charakteristika. Vermutlich modifiziert ProD Menge und Zusammensetzung der Wirtszell-Integrine und verhindert auserdem die Remodulation des Zytoskeletts durch direkte Blockierung von Arp2/3." @default.
