- W2791210357 abstract "Traditionally, metastasis were thought to arise during the late stage of tumour progression. Studies on human cancer, such as breast carcinoma, have shown that cancer cells in a primary tumour exhibit a metastatic phenotype, suggesting that metastatic spread may occur in earlier stages of tumorigenesis. Micro-fabricated devices have started to be employed in recent years to simulate the migration path of cancer cells in vitro and outline the mechanisms of cell motility through confined micro-environments. Previous work suggests that the nuclear envelop can rupture in confinement causing DNA damage and genomic instability, one of the hallmarks of cancer. In this study we observe healthy and cancerous cell lines (MCF-10A, MDA-MB-231, MDA-MB-231 with stable cytosol expressing GFP) with varying motility, adhesion and mechanical properties, migrating in chips equipped with funnel-shaped micro-constrictions. In addition to phase contrast imaging, the cells are also stained with Sir-DNA to track the exact position of the nucleus, and Sir-Actin to enable live cell imaging of the cytoskeletal structure. Absence of nuclear jamming and dissolution of the cytosol are the main requirements for migration through the micro-channels to occur. The cells exhibit different kinds of motility when moving on the chips, such as protrusion-based and bleb-based amoeboid motion. However, they show a repeating migration pattern when making their way through the micro-constrictions. Gaining further insight into the dynamics behind cancer cell migration through confining spaces may help to delineate strategies to restrict the spread of metastatic cells from the primary tumour." @default.
