- W2798815682 abstract "Natamycin is an antifungal agent produced by Streptomyces natalensis, its use as a food additive is mainly related to the surface treatment of cheeses and sausages. The presence of natamycin was evaluated in 138 samples (16 grape juices, 118 wines and four grape and wine derivatives), by the quantitative method of High Performance Liquid Chromatography (HPLC) with Diode Array Detector (DAD). The response of natamycin was not influenced by the effect of the evaluated matrices and the method was linear in the range of work from 0.1 to 5.0 mg L. The detection and quantification limits were, respectively, 0.04 and 0.13 mg L. The absence of natamycin was verified in all samples analyzed by HPLC-DAD. In the confirmation method (qualitative) by HPLC coupled with mass spectrometry (MS/MS) natamycin ionization and fragmentation parameters were considered in order to obtain maximum sensitivity; m/z 503.5 and 485.3 ions were selected to evaluate the presence of this substance on ten samples (seven sweet wines and three grape juice), three of them probably positive (values below the limit of detection by HPLC-DAD). In the best storage conditions tested, sample protected from light and under refrigeration, the initial natamycin concentration (4,96 mg L) in the wine decreased 28% (3,55 mg L) in 60 days." @default.
