- W2799751032 abstract "Lying at the crossroads between sebaceous glands, the infundibulum and the hair follicle, the junctional zone is a region where multiple SC populations have been characterized by the expression of different markers such as Lrig1, Plet-1, Lgr6 and Blimp1. Recently, our lab demonstrated that GATA6, a transcription factor of the GATA family, was also expressed in the junctional zone in mice. There, it is a marker of the sebaceous duct lineage and a regulator of its differentiation. Importantly, by combining genomics, transcriptomics, mouse live imaging, skin reconstitution assays and lineage-tracing experiments, we showed that GATA6-positive cells could dedifferentiate in response to wounding, re-entry a stem cell state and acquire a multi-lineage differentiation potential. This recent work on the role of GATA6 in the mouse upper pilo-sebaceous unit, as well as the high level of conservation of GATA6 during evolution suggests that GATA6 could also be implicated in the maintenance of the upper pilo-sebaceous unit in human. We therefore investigated the expression pattern of GATA6 in human skin. To study the in-depth mechanisms related to GATA6 expression, we designed lentiviral constructs to overexpress GATA6 in human keratinocytes and analysed the differentiation process. Overall, our results suggest that GATA6 is a master regulator of the upper pilo-sebaceous unit homeostasis and differentiation in human." @default.
