- W2801453923 abstract "Mesenchymal stem cells-derived conditioned medium is known to contain various growth factors. Our previous preliminary study revealed that conditioned medium derived from umbilical cord mesenchymal stem cells (UC-MSC CM) that were cultured in hypoxic condition contained high levels of vascular endothelial growth factor (VEGF), a growth factor that stimulates the formation of blood vessels. Therefore, it is hypothesized that UC-MSC CM might substitute growth factor content in endothelial progenitor cells (EPC) culture medium as well as improving the characteristics of either early and/or late EPC. The objective of this study was to investigate the role of UC-MSC-derived VEGF in EPC characterization. Peripheral blood mononuclear cells (PB-MNC) from healthy subjects were differentiated into EPC using EPC culture medium and CM-EPC medium, which was an EPC culture medium added with UC-MSC CM as a replacement to its growth factor component. The surface markers of early and late EPC were observed at the end of culture period using flow cytometry (FACS). Comparison was made from FACS result between EPC that were grown in EPC culture medium and in CM-EPC medium. The present study revealed that the EPC grown in CM-EPC medium had higher expression of specific EPC surface markers, CD31 (early: 67.90%; late: 99.26%) and VE-cadherin (early: 56.69%; late: 64.35%), compared to EPC grown in EPC culture medium (CD31 early: 10.00%—late: 35.63%; VE-cadherin early: 15.58%—late: 11.71%). However, KDR expression level was found low in early and late EPC cultured in both media. The expression of angiogenesis-related genes (VEGFA and CXCR4) in EPC was also studied. It was confirmed that the EPC cultured in CM-EPC medium also expressed both VEGFA and CXCR4. Our findings indicated that replacing growth factor content in EPC culture medium with UC-MSC-derived VEGF could improve characteristics of EPC. This positive effect might be beneficial to develop an alternative composition of culture medium for EPC. Table IEPC culture media composition.
