- W2802250084 abstract "Histological diagnosis is the gold standard to distinguish malignant melanoma from nevus. However, some melanocytic lesions are difficult to diagnose by histological techniques. To solve these problems, some diagnostic tests have been proposed. However, most of them rely on the visual aspects. Therefore, objective methods and numerical data which do not rely on visual aspects are urgently needed. Cyclin D1 gene amplification is a characteristic genetic aberration found in acral melanoma. Using droplet digital PCR assay, we tried to numerical the amplification of Cyclin D1. Formalin-fixed and paraffin-embedded (FFPE) tissue samples of malignant melanoma and nevus were collected. Primary cutaneous melanomas were obtained from 31 patients. Benign nevi were from 15 patients. FFPE sections were sliced into 6-8 μm thick and tumor cells were captured by laser microdissection. Then, the genomic DNA was isolated from collected cells. Amplifications of cyclin D1 were detected by the ddPCR assay by the ratio of cyclin D1( located at 11q13) and housekeeping gene ; adaptor related protein complex 3 beta (AP3B)1 (located at 5q14.1). Mean amplification ratio of cyclin D1 of melanoma group was 1.66(range0.9-8.9,SD=1.45). And that of Nevus group was 1.07(range0.86-1.42,SD=0.16 ). When the cut-off value was set at mean+2SD of the Nevus group (1.38), 22.58%(7/31) of the melanoma were positive while only one nevus showed the positive value(1/15). Amplifications of cyclin D1 detected by ddPCR were verified by fluorescence in-situ hybridization (FISH).The results of amplifications of cyclin D1 detected by ddPCR and FISH were correlated. In conclusion, ddPCR assay for detecting amplifications of cyclin D1 could be a tool of diagnosing malignant melanoma." @default.
