- W2859718140 abstract "Standard tests of stallion semen quality rely on measurements of motility, morphology, and concentration but do not measure the ability of spermatozoa to fertilize. The aim of this study was to develop a method for measuring the binding of stallion spermatozoa to mare oocytes and to relate this to the fertility of a group of commercial stallions. Horse ovaries were collected from an abattoir and subjected to mechanical crushing to release oocytes. An average of 2.1 oocytes per ovary was recovered from 274 ovaries. The oocytes were stored for 6–8 mo in 1 M (NH4)2SO4 before use. Spermatozoa from a control stallion and 12 commercial stallions (7 fertile and 5 subfertile) were labeled with fluorescein isothiocyanate (FITC) and tetramethylrhodamine B isothiocyanate (TRITC), respectively. Equal numbers (35 × 103) of spermatozoa from control and test stallions were added to oocytes in culture at 37°C for 30 min, after which the number of spermatozoa bound to the zona pellucida (ZP) were counted. The sperm-ZP binding ratio (number test sperm bound/number control sperm bound) was 0.77 ± 0.2 (± SEM) for fertile stallions and 0.22 ± 0.1 for subfertile stallions (p = 0.048). The results suggest that the sperm-ZP binding test may be a useful addition to standard methods for assessing stallion semen quality." @default.
