- W286303906 abstract "The protective effect of agents which decrease the oxygen tension in tissue (sodium nitrite and adrenaline) and of other agents which have no effect on the oxygen tension in tissue (cystamine and cysteine) were studied at a normal oxygen content and at a high oxygen content of the inhaled air. It is assumed that an increase in oxygen content of the inhaled air would decrease the effectiveness of sodium nitrite and adrenaline as protective agents, and not affect the effectiveness of cystamine and cysteine. White mice 18 to 20 grams in weight were given a dose of 700 r of Co/sup 60/ gamma rays in hermetically sealed glass containers with a controlled atmosphere of 21% O2 or 98 to 99% O. The protective agent was injected subcutaneously in the following doses per animal before irradiation: 3 mg cysteine, 5 mg cystamine, 0.1 mg adrenaline, and 3.5 mg sodium nitrite. The oxygen tension in the spleen of the animal was determined polarographically. The percent survivals increased from 10.7% for the controls to 32 and 39% survival with the use of cysteine and cystamine as protective agents. The percent survivals did not change, when the irradiation was carried out in pure oxygen.more » The percent survival for adrenaline was 75% on irradiation in 21% O/sub 2/, and the survival dropped to 48.4% on irradiation in pure oxygen. With sodium nitrite the percent survival dropped from 45.2 to 29.9% on going from irradiation in 21% O/sub 2/ to irradiation in pure oxygen. The polarographic data showed an increased content of oxygen in the spleen on increasing the oxygen concentration in the inhaled air. The protective effect of sodium nitrite and adrenaline is associated with a decrease of oxygen in the tissue, but the protective effect of cysteine and cystamine is not related to the concentration of oxygen in the tissue.
