- W2883996965 abstract "Background and Aims: Impaired hepatic autophagy flux (IHAF) is linked with hepatic lipid accumulation (HLA), leading to non-alcoholic fatty liver disease (NAFLD). Here, we analysed the role of Trigonelline (TG), a known anti-oxidant in preventing NAFLD, by modulating the IHAF. Methods: C57BL/6J mice, divided into 4 groups (n = 6), were fed either with Standard Chow or high-cholesterol and high-fat diet (HC-HF), with and without TG (50 mg/kg/thrice a week), for 16 weeks. In-vitro; AML-12 cells and HepG2 cells, were exposed to palmitic acid (PA), with and without TG (25 μM and 50 μM). Cellular events related to IHAF, HLA, lipogenesis and lipo-toxicity were studied by western blotting, immunofluorescence and fluorescence microscopy, and by colorimetric analysis. Results: HC-HF diet fed mice exhibited IHAF, metabolic impairment, HLA, and steatosis. TG administration in HC-HF fed mice, restored hepatic autophagy, along with improvement in metabolic profile, and reduced HLA. AML-12 and HepG2 cells, revealed impaired autophagy, lipid accumulation, and apoptosis, due to PA exposure. TG pre-treatment prevented PA-induced impaired autophagy, which resulted in reduced lipid accumulation. However, the lipid-lowering potential of TG was reduced in the presence of chloroquine and in Atg7 knockdown HepG2 cells, which verify that TG mediates its anti-steatotic effect, mainly via induction of hepatic autophagy. Conclusions: This study proposes a novel therapeutic mechanism of Trigonelline in preventing NAFLD, via restoration of hepatic autophagy in in-vitro and in-vivo settings. Further, clinical studies are warranted to explore Trigonelline, as a therapeutic agent for treating NAFLD. The authors have none to declare." @default.
