- W2885192023 abstract "In current practice of pharmaceutical formulation development, the research and development events are inclined more towards development of physiologically relevant and patient compliance products. Physiological processes affect the performance (absorption, metabolism, elimination, permeation, solubility, and bioavailability) of the administered drug product by any direct or indirect means. The ultimate goal of every pharmaceutical product is to achieve the required bioavailability to get desired clinical effect. Any processing variable or clinical situations including environmental pH, membrane physiology at absorption site that can affect the bioavailability must be considered judiciously during the product development exercises. Based on the widespread knowledge of these physiologic processes, their manipulation can be utilized to further advance the product as well as meet the specific requirement for a market relevant product. This chapter aims to present an extensive discussion to generate basic understanding of subject related matters including physiological processes and various physiological factors affecting product development like EPR effect, environmental pH, first-pass extraction, gastric emptying, gut wall metabolism, route of administration, etc.; along with mathematical models that are relevant for the prediction of physiological processes. A comprehensive account of various transport, metabolism, and efflux modeling procedures is also described. Considering the evolutionary role of computers in the drug discovery and drug design, we have also added a note on the use of computer-based management of physiological processes." @default.
