- W2885330724 abstract "Many plants containing terpenoids are used in traditional medicine for their pain-relieving and antiinflammatory properties. A well-known example is parthenolide, a sesquiterpene lactone produced by superficial leaf glands. While parthenolide is mainly isolated from the feverfew plant Tanacetum parthenium L. (syn. Chrysanthemum parthenium), it can also be found in other species from the Daisy family (Asteraceae species), such as German Chamomile. In European traditional medicine, feverfew has been widely used for the treatment of migraine and rheumatism. It is now recommended as a migraine prophylactic since clinical trials have confirmed its effectiveness against headaches and migraine. Headache has been associated with a predisposed sensitivity of neurovascular reactions to certain stimuli in the central nervous system, with serotonin (5-HT) being the most implicated neurotransmitter in migraine pathophysiology. Drugs that are serotonin antagonists are used in migraine prevention (e.g., methysergide, a semisynthetic ergot alkaloid). It has been suggested that parthenolide is a low-affinity antagonist at 5-HT receptors. The 5-HT receptor is the target of many different therapeutic agents and abused drugs. Recent studies have shown that parthenolide inhibits cocaine-induced behaviors in an animal model, suggesting that parthenolide could have potential use as an overdose antidote or therapeutic agent to treat cocaine intoxication. Cocaine is an addictive drug because of the way that it exploits the dopaminergic neural pathways, and its effect on the body is experienced as a sense of euphoria in human subjects. Cocaine also affects serotonin transporters. Because dopamine and serotonin are both associated with motivation, pleasure, and reward, cocaine is an extremely addictive substance. This review will focus on exploring the properties and mechanisms of action of parthenolide: (1) as a migraine prophylactic, (2) as a cytotoxic/anticancer agent, and (3) as a specific behavioral antagonist of cocaine as a possible treatment for cocaine addiction." @default.
