The goal of this study is to prove the effect of black tea(Camellia sinensis) steepings essence to prevention of the raising of expression of IGF-1 in wistar mouse with highfat diet. The research is conducted by in vivo using rats Wistar strain. Mouse is divided into positive control groupwhich is given high fat diet without black tea steepings essence and treatments group by high fat diet with blacktea steepings essence dose 0,015g, 0,030g and 0,045g/day during 90 day. Expression of IGF-1 is measured by semi quantitative by counting colour intensity at Corell Photo of Paint 11. Intensity expression colour score of IGF-1result of imunohistochemistry are analyzed statistically with One way Anova analysis and analysis of regressionwith level of significancy p = 0,05. Based on the result of analysis at One way Anova (p<0,05) black tea steepingsessence with various dose (0,015g, 0,030g and 0,045g/day) can prevent the raising of expression of IGF-1 significantly(p= 0,000) compared to positive control group. Result of this study prove that black tea (Camellia sinensis)steepings essence can prevent the raising of expression of IGF-1 in Wistar rats with high fat diet ABSTRAK Prevalensi obesitas meningkat di berbagai Negara Asia Pasifik, Eropa, Amerika Serikat, Australia serta di negara-negara berkembang. Perubahan preadiposit menjadi adiposit yang mature memegang peranan dalampatogenesis obesitas. Teh hitam memiliki komponen antioksidan yang banyak berperan dalam pengontrolanproliferasi sel. Theaflavin pada teh hitam kemungkinan dapat menghambat perubahan preadiposit menjadimature adiposit, maka prevalensi terjadinya obesitas sebagai pemicu berbagai penyakit metabolik dapat dicegahsejak dini. Penelitian ini bertujuan untuk membuktikan pengaruh pemberian sari seduh teh hitam (Camelliasinensis) terhadap pencegahan peningkatan ekspresi IGF-1 pada tikus wistar dengan diet tinggi lemak.Penelitian dilakukan secara in vivo dengan menggunakan tikus strain wistar. Tikus dibagi menjadi kelompokkontrol positif dan kelompok perlakuan. Kelompok kontrol positif diberi diet tinggi lemak tanpa sari seduhteh hitam, sedangkan kelompok perlakuan diberi diet tinggi lemak dan sari seduh teh hitam dengan dosis0,015g, 0,030g dan 0,045g/hari selama 90 hari secara bersamaan. Ekspresi IGF-1 diukur secara semikuantitatifdengan menghitung intensitas warna dengan Corell Photo Paint 11. Skor intensitas warna ekspresi IGF-1hasil pewarnaan imunohistokimia dianalisis secara statistik dengan One way Anova dengan taraf signifikansip = 0,05. Berdasarkan hasil analisis One way Anova (p<0,05) sari seduh teh hitam dengan berbagai dosis(0,015g, 0,030g dan 0,045g/hari) dapat mencegah peningkatan ekspresi IGF-1 secara signifikan (p= 0,000)dibandingkan dengan kelompok kontrol positif. Semakin besar dosis teh hitam maka ekspresi IGF-1 makinrendah. Hasil penelitian ini membuktikan bahwa sari seduh teh hitam (Camellia sinensis) mampu mencegahpeningkatan ekspresi IGF-1 pada tikus wistar dengan diet tinggi lemak." @default.
