This chapter reviews the latest achievements in genetic targeting and provides an overview of the benefits of anti-inflammatories, ending with the results from the Cancer Prevention Programme (CaPP). It explores the beneficial effects of aspirin in the context of a high-risk population with mismatch repair gene defects. The identified genetic risk factors lie between the two extremes: rare high-penetrance mutations, which increase the risk for hereditary syndromes, and common genetic variants that confer individually small effects. Aspirin is routinely prescribed as a prophylactic drug for cardiovascular diseases and, with rising evidence for its chemopreventive effect for CRC. Immunological approaches to prevention will be informed by advances in the immunotherapy of existing cancers. Primary prevention strategies, including lifestyle modifications and aspirin use, have been shown to reduce the risk of CRC." @default.
