Pituitary hormones, and ovarian paracrine and autocrine factors regulate expression of the sterol acquisition and steroidogenic machinery by promoting cellular differentiation, and the acute stimulation of hormone biosynthesis by activating the delivery of cholesterol to the inner mitochondrial membrane where the cholesterol side-chain cleavage system resides. This is the rate determining step in steroidogenesis, and it is catalyzed by the steroidogenic acute regulatory protein. The expression of genes involved in cholesterol biosynthesis, lipoprotein uptake, intracellular cholesterol trafficking, and steroid synthesis is controlled by an overlapping set of transcription factors that function in a combinatorial fashion. This chapter reviews the essential components of sterol metabolism and the steroidogenic machinery in the ovary, focusing on the initial steps in steroid hormone biosynthesis, the acquisition of the steroid hormone precursor, cholesterol from circulating lipoproteins or de novo biosynthesis, and its metabolism into pregnenolone, the first committed step in synthesis of steroid hormones." @default.
