In addition to supporting first-in-human studies in compliance with ICH S7A and S7B, cardiovascular safety pharmacology studies can be deployed throughout the drug discovery and development stages. The impact of such studies includes, but is not limited to, providing confidence in the target, influencing the structure–activity relationship, contributing to the design or resumption of clinical trials, assessing drug–drug interactions, or supporting marketed products. Four case studies are presented in this chapter. The generation of hypothesis-driven mechanistic cardiovascular safety studies, their careful execution, analysis, interpretation, and contextualization enabled (1) to resume ZD6126 clinical trials, (2) to progress AZD7762 into the clinic with appropriate cardiac monitoring, (3) to support the postapproval commitment toward patients prescribed with hydroxyzine, and (4) to provide confidence in the safety of levocetirizine and ensure optimal clinical monitoring. Those examples illustrate the influence and impact of cardiovascular safety pharmacology studies on drug discovery and development." @default.
