- W2891689541 abstract "Restricted endonucleases are enzymes that help in identifying nucleotide sequences specifically in double-stranded DNA. This type of enzyme works naturally as a restriction for prokaryotic cells. It breaks foreign DNA that enters the bacterial cell, but it does not remove the host DNA that has been protected by methylation. The restricted endonucleases naturally cleave DNA at a precise point within a recognition sequence. For molecular cloning techniques, this precision is needed mainly for separating genes from genomic DNA or inserting foreign DNA into a plasmid. This chapter explains the way in which a large number of DNA fragments can be generated by restriction enzymes and with the use of agarose-gel electrophoresis, it can be shifted from one place to the other. While the movement is possible, it has to be understood that the negatively charged DNA molecules are drawn to the anodes only when they are under the influence of an electric field. To determine the size and purity of both maxi-prep and mini-prep, restriction enzymes and agarose mini-gels are used as quick diagnostic tools." @default.
