- W2892110670 abstract "Publisher Summary 6-Phospho-D-gluconate is formed in Pseudomonas by the dehydrogenation of D-glucose 6-phosphate and subsequent hydrolysis of 6-phospho-D-glucono-l,5-1actone and by the hydrogenation of 2-oxo-6-phospho-D-gluconate. It is metabolized either by the 6-phospho-D-gluconate dehydratase in the Entner–Doudoroff pathway, or by the 6-phospho-D-gluconate dehydrogenase in the hexose monophosphate pathway. The assay is based on the spectrophotometric determination of nicotinamide adenine dinucleotide dehydrogenase (NADH) formed with 6-phospho-D-gluconate as a substrate at high pH conditions. The steps involved in the purification of 6-phospho-D-gluconate are (1) the preparation of cell-free extract, (2) bulk separation on CM-Sephadex, (3) ammonium sulfate precipitation, (4) diethylaminoethyl (DEAE)-cellulose chromatography, (5) hydroxyapatite chromatography, and (6) preparative electrophoresis. Lyophilized bacteria are used as the enzyme source. All solutions are 20 m M in 2-mercaptoethanol and all operations are performed at 0–4°, unless otherwise stated. The purified enzyme with a specific activity of 121 units/mg is homogeneous by disc gel electrophoresis, by sodium dodecyl sulfate electrophoresis, and by gel isoelectric focusing." @default.
