- W2892897068 abstract "Abstract Epigenetic modifications acquired during oocyte growth are essential for transcriptional regulation, establishment of maternal DNA methylation imprints, and the maintenance of chromosome stability. Human and mouse oocytes exhibit a unique nuclear organization that is subject to dynamic changes in a large-scale chromatin structure regulated by chromatin remodeling proteins and histone posttranslational modifications. Functional differentiation of chromatin structure or epigenetic maturation confers the oocyte with full meiotic and developmental potential. Establishment of DNA methylation occurs on a locus-by-locus basis, and both histone modifications as well as transcription at regulatory sequences dictate the placing and timing of methylation marks at specific genes. The presence of germ cell-specific epigenetic modifications may also be vital for the oocyte-to-embryo transition. Importantly, large-scale chromatin remodeling is indispensable for meiotic centromere function and accurate chromosome segregation. Yet, these key mechanisms are susceptible to environmental disruptions and reproductive senescence, leading to increased oocyte aneuploidy. Understanding the regulation of epigenetic maturation is crucial to determining the effect of environmental or hormonal disruptions on oocyte quality and to uncover the epigenetic origins of aneuploidy." @default.
