- W2892955547 abstract "Publisher Summary This chapter discusses the poikilotherm cell identity by cytotoxic antibody test. The use of poikilothermic species as necessary tools for research in viral diseases, molecular biology, and oncology of cold-blooded vertebrates increased the need for constant surveillance and characterization of these cultures in the laboratory to assure such freedom from contamination. Most amphibian, reptilian, and fish cell lines can be grown in culture media suitable for mammalian cells. Antisera are prepared by immunizing rabbits with muscle tissue from fish, amphibians, and reptiles. Absorption of antisera is sometimes required to ensure specificity. The various poikilothermic and homeothermic animal antisera, 0.1 ml, to be tested are added to the tubes and the cell–antiserum mixtures incubated at 37°C for 1 hour. Fresh guinea pig serum can be ampoulized and stored in liquid nitrogen at −196°C for a minimum of 2 years or lyophilized and reconstituted for the test with sterile distilled water. Approximately, 0.1 ml of 0.25% trypan blue in phosphate-buffer saline (pH 7.2) is added to the tubes and samples of each cell–serum specimen are placed in a hemocytometer and the stained (killed) and unstained (viable) cells counted. A cell control containing normal rabbit serum with and without complement must be included in the test." @default.
