- W2894615148 abstract "Horizontal gene transfer (HGT) is a central evolutionary process enhancinggenome diversification and rapid adaptation of species to new environmentalconditions. Mobile genetic elements (MGE) facilitate genetic exchange betweenspecies through HGT by carrying accessory gene cargos encoding beneficial traitssuch as resistance to metals and antibiotics. MGE-mediated transfer of antibioticresistance genes between species in natural microbial communities hascontributed to the global spread of antibiotic resistance. It is therefore essentialto understand the ecological drivers of the maintenance and transmission ofMGEs in bacterial communities. Here I use conjugative plasmids as an exampleMGE to study the ecological and evolutionary dynamics of plasmids in bacterialpopulations and communities across a range of environments. First, Idemonstrate that plasmids selected in a single-host environment evolved hostspecialism due to fitness trade-offs, whereas plasmids evolved in a multi-hostenvironment could overcome this trade-off to evolve host-generalism. Secondly, Ishow that the costs and benefits of plasmid carriage and the long-term dynamicsof the plasmid and the mercury resistance transposon it encodes variedextensively between diverse species of Pseudomonas. I next show that plasmidmaintenance was facilitated by compensatory evolution to ameliorate the cost ofplasmid carriage. Compensatory loci varied between species, with parallelmutations targeting different regulatory and biosynthetic pathways in eachspecies. Lastly, I examine the effect of community structure on plasmiddynamics in simple bacterial communities. When plasmids were carried byproficient plasmid-donor species this led to higher plasmid abundance at thecommunity-level, while in diverse communities, plasmid transmission could beimpeded through the dilution effect, limiting plasmid spread. This thesisdemonstrates that plasmid dynamics in bacterial communities are determined bythe combination of ecological and evolutionary processes, depending on theselective environment, the structure of the bacterial community and variationamong species in their proficiency to host plasmids and to undergo compensatoryevolution to ameliorate their costs. These data highlight the importance ofstudying plasmid dynamics in a community-context." @default.
