- W2894750280 abstract "SESSION TITLE: Lung Cancer SESSION TYPE: Original Investigation Posters PRESENTED ON: 10/10/2018 01:00 PM - 02:00 PM PURPOSE: Epidermal growth factor receptor (EGFR) mutation test is very important for selecting chemotherapy regimen in patients with advanced lung cancer. Tissue biopsy sample is standard material for this, but there are several limitations. Existing methods for plasma test for EGFR mutation has low sensitivity. We tested feasibility of peptide nucleic acid (PNA) clamping method for EGFR mutation. METHODS: Thirty-nine patients with newly diagnosed, never treated lung cancer were enrolled. We tested EGFR mutation in both tumor tissue and plasma samples. The circulating tumor DNA (ctDNA) was extracted from the plasma and analyzed by two different methods of PNA clamping-assisted fluorescence melting curve analysis (PNAc) and droplet digital PCR (ddPCR) for EGFR mutation. Both were compared with the results of tumor tissue. RESULTS: EGFR mutations including exon 18, 19, 20 and 21 were detected in 19 cases (48.7%) by PNAc and 16 cases (41.0%) by ddPCR in 39 cases. Using EGFR mutation status of tumor tissue as a standard reference, the sensitivity and specificity for PNAc were 60.0% and 88.9% and the sensitivity and specificity for ddPCR were 53.3% and 100% respectively. The concordance rate between two methods was 82.1% (32/39). Seven cases showed discrepant results between two methods, of these cases 4 cases analyzed by PNAc were matched to tumor tissue results, whereas 3 cases analyzed by ddPCR ware matched to tumor tissue results. CONCLUSIONS: Plasma EGFR test by PNAc is sensitive method compared with ddPCR, and concordance rate with ddPCR was high, so it is feasible for the large number case tests.
