- W28955079 abstract "Neurotrophins regulate the generation, differentiation, survival, and maintenance of vertebrate neurons by interacting with two classes of receptors. The low-affinity neurotrophin receptor (p75LNGFR) is a transmembrane glycoprotein initially isolated by its ability to bind nerve growth factor (NGF); however, subsequently it is shown to bind all the members of the neurotrophin family with equal affinity. The low-affinity neurotrophin receptor, p75LNGFR, is structurally related to the tumor necrosis factor receptor and to CD40 and it contains a short cytoplasmic domain of unknown biochemical function. This receptor mediates activation of the sphingomyelin cycle by NGF. This chapter describes the results of investigations performed to elucidate the structural requirements for binding to p75LNGFR in brain-derived neurotrophic factor, NT-3, and NT-4 using chimeric and mutant neurotrophins generated by site-directed mutagenesis. The chapter also describes the similarities and differences in the way neurotrophins interact with p75LNGFR." @default.
